Since then, excellent progress has been made on the study of the mechanisms of poly(ADP ribose) reaction. The last five years have been particularly exciting since the development of various molecular biology techniques has revealed the complex nature of this multifunctional enzyme.
The poly(ADP-ribose) polymerase (PARP) protein family generates ADP-ribose (ADPr) modifications onto target proteins using NAD + as substrate. Based on the composition of three NAD + coordinating amino acids, the H-Y-E motif, each PARP is predicted to generate either poly(ADP-ribose) (PAR) or mono(ADP-ribose) (MAR).
However, the reaction product of each PARP has not been clearly. Post-translational modifications of cellular proteins by mono- or poly-ADP-ribosylation are associated with numerous cellular processes. ADP-ribosylation reactions are important in the nucleus, and in mitochondrial activity, stress response signaling, intracellular trafficking, and cell senescence and apoptosis decisions.
